Curcumin and its analog alleviate diabetes-induced damages by regulating inflammation and oxidative stress in brain of diabetic rats

Abstract Background Diabetic encephalopathy is a severe diabetes complication with cognitive dysfunction and neuropsychiatric disability. The mechanisms underlying diabetic believed to be relevant oxidative stress, vascular amylin deposition, immune receptors, inflammation, etc. This study wanted evaluate the ability of curcumin its analog A13 alleviate stress inflammation in diabetes-induced damages brain. Methods Sixty adult male Sprague–Dawley rats were divided into 5 groups: normal control (NC) group, mellitus (DM) curcumin-treated (CUR) high dose A13-treated (HA) low (LA) group. Activation nuclear factor kappa-B (NF-κB p65) pathway was detected by RT-qPCR, immunohistochemical (IHC) staining Western blot; biochemical detection kit; brain tissue sections stained hematoxylin–eosin (HE) Myelin staining. Results IHC blot showed that treatment could inhibit NF-κB p65 pathway. Curcumin increased activity superoxide dismutase decreased malondialdehyde level rats. Furthermore, HE demonstrated histological lesions significantly ameliorated A13. Conclusion regulating pathways stress. may new potential therapeutic agent for encephalopathy.
